چکیده

Introduction Neonatal Onset Multisystem Inflammatory Disorder (NOMID) is the most severe of the Cryopyrin-Associated Periodic Syndromes (CAPS) spectrum [1]. Given the persistent nature of CAPS, symptoms may be confused with autoimmune diseases. However, unlike autoimmune diseases, autoinflammatory disorders are not associated with high-titer autoantibodies or antigen-specific T cells. Here we describe a patient with severe and early onset features of NOMID confirmed by NLRP3 mutation, hepatitis, neonatal antibodies and a limited response to high dose anakinra.

PW02-039 - Long-term anakinra treatment in CAPS: a metaanalys

Introduction The common denominator in CAPS (FCAS, MuckleWells syndrome, NOMID/CINCA) is an uncontrolled IL-1b release. An often complete response after treatment with the IL-1 blocker anakinra (Kineret) has been demonstrated in all three entities of CAPS [1-3]. However, the overall documentation is limited due to the inherent difficulties in conducting randomized studies in the more severe for...

PW02-028 - Association of novel NLRP3 mutations with CAPS phenotype in Turkish patients

Introduction Cryopyrin-Associated Periodic Syndromes (CAPS) are a group of rare, inherited, autoinflammatory diseases involved of Familial Cold Autoinflammatory Syndrome (FCAS), Muckle-Wells Syndrome (MWS) and Neonatal Onset Multisystem Inflammatory Disease (NOMID) (also called Chronic Infantile Neurologic Cutaneous Articular, or CINCA, Syndrome. The responsible gene NLRP3 (nucleotide-binding d...

PW02-029 - Single cell fluorescent immunoassay of CINCA/NOMID

Introduction CINCA syndrome, also known as NOMID, is a rare autoinflammatory disease caused by the NLRP3 mutations. It has been known that conventional genetic analysis failed to detect disease-causing mutations in approximately 40% of patients. We have recently identified NLRP3 somatic mosaicism on 70% of these ”mutation-negative” patients in the international collaborative study (Tanaka N. an...

PW02-033 - Cytokine profile in CSF in CAPS patients

Introduction CAPS is a rare autoinflammatory syndrome caused by autosomal dominant mutations in the NLRP3/CIAS 1 gene on chromosome 1q44 encoding for the cryopyrin protein, an important component of the inflammasome, leading to excessive production of interleukin-1beta (IL-1ß). CAPS encompasses three different entities of variable clinical severity: familial cold auto-inflammatory syndrome (FCA...

Understanding the pathophysiology of NOMID arthropathy for drug discovery by iPSCs technology

Introduction and objectives NOMID, also known as CINCA syndrome, is a dominantly inherited autoinflammatory disease caused by NLRP3 mutations. The pathophysiology of NOMID is explained by gain of function mutation of NLRP3, which activates NLRP3 inflammasome and produce an excess of IL-1b. This mechanism is supported by clinical observation that anti-IL-1 therapy is effective on its systemic in...
